Three test kits to examine morphology and vitality of sperm cells

Semen Stain is a quick staining method to assess the morphology of spermatozoa (spermiogram) and allows differential staining of the different sperm parts due to their specific basophilic, eosinophilic and neutrophilic properties.

The sperm cells are fixed. Here, the succedan-staining technique is used, which means, three dyes are used one after the other and it results in differential staining of different tissues with individual dyes.

  • Morphology assessment of spermatozoa using succedanstaining method
  • Differential staining of spermatozoa tissues based on their basophilic, eosinophilic and neutrophilic properties

The SemenVit kit is performed to examine the vitality of sperm cells. It is particularly important in semen samples with less than 40% of forward moving (motile) sperms.

The determination of the vitality of sperm cells is judged by the integrity of the sperm membrane. The dye exclusion method is based on the fact that dead sperms with damaged plasma membranes absorb certain dyes.

  • Vitality assessment of spermatozoa using dye exclusion method.
  • Based on integrity of the sperm membrane in live spermatozoa.

This SemenHos test is used to test the vitality of sperm cells. The hypo-osmotic swelling is based on the semi-permeability of the intact cell membrane and their ability of active water transport in order to not burst. In sperm cells with intact membranes the flagellum swells up within 5 min. This change remains stable up to 30 min.

In this hypo-osmotic swelling test swelling of cells only occurs in vital cells with an intact membrane by using a hypotonic solution.

  • Vitality assessment of spermatozoa using the hypnos-motic swelling method.
  • Based on the semi-permeability of the intact cell membrane and the possibility of active water transport
  • Swelling effect in intact sperm cells can be observed for up to 30 minutes
